The Klingon-Romulan border was the border between the Klingon Empire and the Romulan Star Empire in the Beta Quadrant.

Camp Khitomer was located near the border on the Klingon side. (Star Trek VI: The Undiscovered Country)

In early 2368, Jean-Luc Picard placed a fleet of twenty-three Federation starships, designed to establish a tachyon detection grid along the Klingon-Romulan border in an attempt "to prevent any external power from interfering in Klingon affairs." According to Commander Sela, "deploying an invasion fleet along our border is a clear act of aggression. [..] It will not be tolerated." She then gave Picard "twenty hours to recall your ships and return peacefully to Federation territory." (TNG: "Redemption II")

In early 2372, the Klingon Empire attacked several Romulan outposts along the border, in an attempt to "[reassert] themselves all over the quadrant." (DS9: "Hippocratic Oath")

It is implied in "Hippocratic Oath" that this border extended into the Alpha Quadrant; however, the star chart seen in Star Trek: Picard would seem to argue against this, placing where the border where it would have been squarely in the Beta Quadrant.
It is implied by the star chart seen in the Star Trek: Picard episode "Maps and Legends", that by 2399, the border had ceased to exist as there were no shared borders between the Klingon Empire and the Romulan Free State, the successor to the Romulan Empire, and where there was a border there was now neutral space.
